TEST İLKESİ
| 本试剂盒は競合ELISA法を採用しています。マイクロプレートにねずみ GHRH抗原を固相化し、検体または標準品中のねずみ GHRHが固相化されたねずみ GHRHと、ビオチン標記抗ねずみ GHRH単クローン抗体の結合部位を競合します。遊離した成分は洗浄により除去し、次いでHRP標記アビジンを添加してビオチン-アビジン特異的結合を形成させ、再び遊離成分を洗浅除去します。発色基質（TMB）を添加するとHRPの触媒により青色に呈色し、反応停止液を加えることで黄色に変化します。マイクロプレートリーダーで450 nmの吸光度（OD値）を測定し、ねずみ GHRH濃度はOD450値と逆相関することから、検量線を作成して検体中のねずみ GHRH濃度を算出します。 |
