TEST İLKESİ
| 本試験キットは、サンドイッチELISA法を採用しています。酵素標識プレートに抗ねずみ TPPP抗体を固相化し、検体（または標準品）中のねずみ TPPPが固相化抗体と結合します。その後、ビオチン標識抗ねずみ TPPP抗体とHRP標識アビジンを順次添加すると、ビオチン標識抗体は固相化抗体に捕捉されたねずみ TPPPに結合し、ビオチンとアビジンが特異的に結合して免疫複合体を形成します。遊離成分は洗浄により除去され、発色基質（TMB）を添加するとHRPの触媒作用により青色に呈色し、反応停止液を添加することで黄色に変化します。マイクロプレートリーダーで450 nmの吸光度（OD値）を測定し、ねずみ TPPP濃度とOD450値の間に正の相関が認められるため、標準曲線より検体中のねずみ TPPP濃度を算出します。 |
