TEST İLKESİ
| 本試験キットは、サンドイッチELISA法を採用しています。酵素標識プレートに抗ねずみ PDL1抗体を固相化し、検体（または標準品）中のねずみ PDL1が固相化抗体と結合します。その後、ビオチン標識抗ねずみ PDL1抗体とHRP標識アビジンを順次添加すると、ビオチン標識抗体は固相化抗体に捕捉されたねずみ PDL1に結合し、ビオチンとアビジンが特異的に結合して免疫複合体を形成します。遊離成分は洗浄により除去され、発色基質（TMB）を添加するとHRPの触媒作用により青色に呈色し、反応停止液を添加することで黄色に変化します。マイクロプレートリーダーで450 nmの吸光度（OD値）を測定し、ねずみ PDL1濃度とOD450値の間に正の相関が認められるため、標準曲線より検体中のねずみ PDL1濃度を算出します。 |
